I currently have a Fulsac header and 2 inch baffles on my bike and a buddy has a 2 into 1 that I want to try. Is there enough of a difference in exhaust to warrant a retune?

 Yes, it would require a retune.  Unless you have a radical combo, you will not gain much if any from your current exhaust.  The Fullsac never disappoints.

If all you want to do is try out the other exhaust, you should be able to do so without a new tune.  If you decide to keep the new exhaust, then I'd spend the money to have the tuner tweak your tune.
