this may sound like a stupid question.  Is there a certain gear that a bike is put in to do a dyno pull? 1:1?  and if so is there ever a problem with wheel spin on the drum with Harley bikes?  On sport bikes we would do several pulls cause this would happen and also to make sure that we was getting the same reading out of the engine as well.  also.  we also would post vehicle speed.  I dont think any one here really cares about mph. which is ok with me. dont race this bike.   but just wondering.

Whatever gear that does not use any of the gearbox's mechanical leverage, plus or minus.

It is a Great question.  I have found not everyone runs the Dyno's the same which makes comparing the numbers..questionable.
